Greater number of mesenchymal stem cells released with fibrous synovium in culture model

Investigators of this study found fibrous synovium compared with adipose synovium release a greater number of mesenchymal stem cells in a suspended synovium culture model.
Researchers harvested human synovial tissue from knee joints during the total knee arthroplasty of 34 patients with osteoarthritis. Investigators suspended 1 gram of synovium with a thread in a bottle filled with 40 mL of culture medium. A 3.5-cm diameter culture dish was at the bottom of the bottle and investigators examined the culture dish after 7 days. Multipotentiality and surface epitopes were analyzed for the cells harvested. Investigators compared the number of colonies derived from the fibrous synovium and the adipose synovium.
In all 28 knees, investigators saw colonies of spindle-shaped cells. Investigators noted, on average, colonies numbered 26 and multipotentiality for chondrogeneis, adipogenesis, calcification and surface epitopes similar to mesenchymal stem cells were seen in the cells derived from colony-forming cells.
