Leukocyte-rich PRP, red blood cell-treated synovial cells experience significant cell death

Significant cell death and proinflammatory mediator production resulted when synovial cells were treated with leukocyte-rich platelet-rich plasma and red blood cell concentrate, according to study results.

Researchers obtained and processed peripheral blood from four donors to create leukocyte-rich platelet-rich plasma (LR-PRP), leukocyte-poor platelet-rich plasma (LP-PRP), red blood cell concentrate and platelet-poor plasma and cultured them with human synoviocytes for 96 hours using standard laboratory conditions. The researchers then evaluated cell viability and inflammatory mediator production.

Synoviocyte death was significantly greater with LR-PRP than with LP-PRP, phosphate-buffered saline and platelet-poor plasma, according to the researchers. Synoviocytes also demonstrated significantly greater cell death when treated with red blood cell concentrate compared with phosphate-buffered saline, platelet-poor plasma, LP-PRP and LR-PRP.

Interleukin (IL)-1β and IL-6 content were both significant higher in cultures treated with LR-PRP compared with all other culture conditions. The researchers also found the greatest tumor necrosis factor-alpha levels in LP-PRP, whereas interferon-γ levels were greatest in red blood cells than in any other culture condition.
